Dynabeads Intact Virus Enrichment beads are 1-μm strong anion exchange magnetic beads that bind negatively charged vesicles and molecules. The beads can enrich for enveloped viruses such as, but not limited to, SARS-CoV-2 coronavirus, human immunodeficiency virus (HIV), respiratory syncytial virus (RSV), norovirus, adenovirus, enterovirus, and Ebola-, Zika-, Influenza A- viruses.
The enrichment is gentle and fast and can be performed in as little as 15 minutes. The virus can be easily removed from the beads in an additional 10-minute release step.
The Dynabeads Intact Virus Enrichment beads can also be used to isolate extracellular vesicles (EV’s) and exosomes with the same rapid and efficient enrichment and release method. Successful enrichment of exosomes has been performed from cell culture media and liquid biopsy samples such as serum, plasma and urine. In addition, the Dynabeads magnetic beads allow for high-throughput enrichment in combination with any of our KingFisher instruments, thus both manual and automated protocols are available.

Downstream applications
The enriched intact virus can be used for further virus growth or in any downstream functional assays, protein research (western, mass spec), purification of RNA for PCR, etc. In addition, virus-like particles (VLPs), containing only virus-specific proteins and not nucleic acids, are extensively used in viral proteomic research. Dynabeads Intact Virus Enrichment beads isolate both live and inactivated intact viruses and VLPs.
Dynabeads Intact Virus Enrichment beads also isolate EV’s and exosomes and can, in addition to the applications above, be detected by flow cytometry using the Dynabeads-based Exosome-Human CD81 Flow Detection Reagent, for example, or the equivalent Dynabeads-based products for CD9 or CD63. For analysis of other targets, the Dynabeads-based Exosome-Streptavidin Isolation/Detection Reagent can be used in combination with biotinylated detection antibodies.
Easy and efficient protocol
After a quick wash of the beads prior to use, the Dynabeads Intact Virus Enrichment beads are added to the sample in a short 10-minute incubation step. Wash the bead-bound virus once on a DynaMag magnet to remove the unbound target and add an appropriate buffer for 10 minutes according to the manual (below) if release from the beads is required. This quick and efficient isolation of viruses and EV’s is an improvement from the traditional and tedious ultracentrifugation methods that also require large and expensive equipment and give questionable yield.

The Dynabeads magnetic beads are ideal for high-throughput enrichment. This process can be automated using any of our KingFisher sample purification systems or liquid handlers.
