Agar is a mixture of polysaccharides extracted from red seaweed cell walls. It has unique gelling properties that are utilized in many industrial and laboratory applications. Solid culture medium is created by adding agar to media broth (typical final concentration is 1–2%). Solid media cell culture allows the isolation of individual E. coli colonies and is fundamental in cloning workflows.
Agar is used in diagnostic laboratories to detect pathogens from contaminated food or water. Also, it can be used to prepare selective media for saccharomyces cerevisiae and other yeasts used in various molecular microbiology procedures (e.g., YPD Agar). In addition, agar has been used in tissue double embedding for histopathology, hydrogel creation, and tissue engineering applications.
Primary applications
• Making solid media to grow cells (e.g., bacteria, fungi, yeast)
• To isolate E. coli clones with plasmid on a selective media
• Tissue engineering, hydrogel production
