The QuantiGene Plex Human Akt Signaling Pathway Panel, 12-plex, enables measurement of expression of relevant human genes associated with the Akt signaling pathway through the analysis of 12 RNA targets and housekeeping genes in a single well using Luminex xMAP technology in combination with branched DNA signal amplification. This panel contains genes that are involved in signaling pathway dynamics and this pathway’s role in cellular processes such as cell survival, proliferation, metabolism, and apoptosis.
QuantiGene Plex panels are extensively tested for target combinability, interference, and cross-reactivity to provide the outstanding levels of validation and precision.
This panel is used in combination with the QuantiGene Plex Assay Kit, which contains all buffers, plates, and detection reagents necessary to perform the assay in 96-well plates.
Features of QuantiGene Plex panels include:
• True multiplexing—measure multiple genes of interest and housekeeping genes in the same well with no cross-reactivity
• Accommodation of difficult sample types—works with degraded and cross-linked RNA in FFPE tissues and blood
• Standardized platform—uses 96-plate format compatible with Luminex 200, FLEXMAP 3D, and INTELLIFLEX systems (384-well format available on demand)
• Simple workflow—employs an ELISA-like workflow with direct hybridization of transcripts to beads and transcript labeling (branched DNA signal amplification and PE readout)
• Customize with validated targets—if alternative targets are needed in this panel, select from our large inventory of over 22,000 genes to create custom research panels
• Rapid turnaround of this catalog panel for fast delivery
Targets: IGF1R, IRS1, AKT1, AKT1S1, GSK3B, MTOR, RPS6KB1, CREB1, GAPDH, HPRT1, PPIB, GUSB
Panel and signal amplification technology
QuantiGene Plex assays are hybridization-based assays for the direct quantitation of RNA directly from cell lysates, tissue homogenates, or purified RNA, and they enable the measurement of multiple targets in the same well via multiplexing. QuantiGene Plex panels contain two components: a target-specific probe set and magnetic capture beads. The target-specific probes are used to capture the RNA of interest, and branched DNA technology then amplifies the fluorescent signals, which are read with a Luminex xMAP instrument. The intensity of the fluorescent signal is directly proportional to the amount of target mRNA present in the sample. This allows for (relative) quantification of gene expression and, with this specific panel, enables human immune profiling. Detailed and step-by-step data analysis is facilitated by the QuantiGene Plex data analysis app.
Applications
QuantiGene Plex assay research applications include compound screening of drug candidates, biomarker verification, siRNA knockdown efficiency determination, prospective and/or retrospective analysis of clinical samples, microarray validation, predictive toxicology research, and translocation/fusion gene detection. In general, any multiplex gene expression analysis can be done with QuantiGene Plex assays, independent of species or sample type.
ProcartaPlex protein testing—one machine, two applications
Gain deeper insight with complementary and highly correlative gene expression and protein assays. The ProcartaPlex Human Akt Pathway Dual Reporter Panel, 2x8plex (Cat. No. EPX080-97200-DR) enables simultaneous quantitation of total and phosphorylated proteins involved in signaling pathway dynamics.
